Typical adult replacement doses delivered by HGH pens: 150–500 mcg/day (0.15–0.50 mg/day), titrated individually; some patients start lower (e.g., 50–100 mcg/day) and some regimens for other uses go higher. Humatrope® (somatropin) is typically administered as a subcutaneous injection, with dosages individualized based on the patient's weight and specific condition. For adults with growth hormone deficiency, the usual starting dose is 0.006 mg per kg of body weight daily, which may be adjusted by the doctor based on the patient's response to treatment.

Humatrope is available in three cartridge strengths: 6 mg, 12 mg, and 24 mg, and each cartridge must be used with the corresponding HumatroPen size to ensure proper dosing.

Saizen HGH Protocol - Typical AGHD Protocol

Starting dose (weight‑based): ≤0.005 mg/kg subcutaneously once daily; may increase to ≤0.01 mg/kg/day after ~4 weeks, titrate by clinical response and age/gender‑adjusted IGF‑1. Alternatively if non‑weight consideration for protocol, start ~0.15–0.30 mg/day and commonly ~0.2 mg/day; increase by ~0.1–0.2 mg/day every 1–2 months as needed.  Older or obese patients with BMI >30: start lower and use smaller increments; Women, especially on oral estrogens often require higher HGH doses.  Typical evaluation period is initial titration for evaluation over ~3–6 months with reassessment of symptoms and IGF‑1 levels, continue long‑term if beneficial.

Preparation (vials): reconstitution guidance using 5 mg vial with 1–3 mL bacteriostatic water or 8.8 mg vial with 2–3 mL bacteriostatic water (expect ~10% mechanical loss with multi-dose administration). Use appropriately sized syringes or pen cartridges. Administration: subcutaneous injection, often in evening; rotate injection sites.

Saizen Devices include Easypod® (electronic auto‑injector), Aluetta® (mechanical pen with Smartdot™); SaizenPrep® a reconstitution system for cartridges vs vials. Cartridge sizes are commonly supplied as 4 mg, 5 mg, 8.8 mg, and 12 mg. HGH vials can be 5 mg and 8.8 mg.

Using a Vial and Bacteriostatic Water for Administration

Storage: Store in the refrigerator BEFORE AND AFTER MIXING.

Special Considerations: Rotate injection sites. Kit comes with a vial of Omnitrope in powder form and a vial of liquid diluent for bacteriostatic injection (1.14mL total).

Supplies: 22 gauge 1 inch or 1 ½ inch needle plus a 3 mL syringe to dilute/mix Somatropin, BD insulin syringe microfine 28 gauge ½ inch 0.5mL to administer Somaropin, alcohol swabs.

Instructions

1. Wash your hands and work on a clean, dry surface.

3. Attach a 22 gauge 1 ½ inch needle to a 3 mL syringe.
4. Remove the protective caps form both vials. Clean the tops of both vials with an alcohol swab.
5. Pull back the plunger to draw up ~1 mL of air into the syringe.
6. Insert the needle into the vial of bacteriostatic diluent and push the air into the vial.
7. Turn the vial upside down with the needle still in the vial. Adjust the needle so that the tip of the needle is in the liquid and not the air space. Slowly pull back on the plunger to draw up 1.14 mL (the whole vial) of bacteriostatic diluent. If there is air in the syringe, push the liquid back into the vial and withdraw again. You only need to worry about air bubbles that extend from one side of the syringe to the other, any bubble smaller than that is nothing to worry about.
8. Remove the needle form the vial of bacteriostatic diluent.
9. Inject the bacteriostatic diluent in the syringe into the vial of HGH powder.
10. Swirl the vial gently until all powder is dissolved.
11. Using an insulin syringe (syringe with orange cap), draw up 25 units.

Administration (Subcutaneous)

1. Choose an injection site on your abdomen or hip area.
2. Clean the area with an alcohol swab and allow to dry.
3. Pinch up the skin.
4. Insert the needle straight into the skin- do not go in at an angle.
